The study analyzed approximately 18,000 people with Huntington's disease to identify genetic factors influencing the onset of psychiatric, motor, and cognitive symptoms. Genome-wide analysis confirmed 11 previously known modifiers of motor onset and discovered a novel genetic locus spanning RAB3B and ZFYVE9 genes associated with the age of violent behavior onset. The research found that certain genetic variants, such as those in MSH3, FAN1, and HTT genes, influence both psychiatric and motor symptom onset. Additional variants in PMS1 and POLD1 genes were associated with motor symptoms. Psychiatric polygenic scores successfully predicted psychiatric symptom onset, suggesting a combined genetic architecture linking general psychiatric liability with pathways specific to Huntington's disease and repeat expansion disease.
