The TRF2 protein, previously known mainly for protecting chromosome ends, is also crucial for muscle stem cells. It helps these cells maintain their identity and coordinate the cycle of rest, repair, and renewal of muscle tissue. Researchers at the Perelman School of Medicine at the University of Pennsylvania found that without TRF2, muscle stem cells lose their identity. Instead of repairing damaged muscle, fat and scar tissue accumulate in the affected areas. The study, published in Science Advances, also showed that removing TRF2 in mice with Duchenne muscular dystrophy significantly accelerated disease progression. TRF2 also acts in regulatory regions throughout the genome, not just at telomeres, thereby preserving the regenerative capacity of muscle stem cells. This discovery could lead to new therapeutic approaches for muscular dystrophy and provide insights into cancer biology.
