RNA Splicing of the REST Gene by SRRM4 in Vascular Smooth Muscle Cells Promotes Abdominal Aortic Aneurysm Development

Abdominal aortic aneurysms (AAA) are associated with changes in the phenotype of vascular smooth muscle cells. Researchers identified the SRRM4 protein as a key regulator of AAA development through analysis of genes in human and mouse tissues affected by aneurysm. When mice lacking SRRM4 specifically in vascular smooth muscle cells were exposed to aneurysm-inducing stimuli, there was a significant reduction in aneurysm development. SRRM4 affects cell phenotype through alternative splicing of the REST gene, leading to the production of a non-functional REST4 variant. The research further showed that REST directly suppresses the STAT1 protein, and that STAT1 inhibition was beneficial in preventing AAA and slowing disease progression in mouse models. These findings suggest a novel potential target for developing therapeutic strategies for abdominal aortic aneurysms.
