Evorpacept plus trastuzumab, ramucirumab and paclitaxel in HER2-positive gastric cancer: a randomized phase 2 trial

This study focused on the phase 2 portion of the randomized phase 2/3 ASPEN-06 trial. It involved patients with advanced HER2-positive gastric cancer whose disease progressed despite prior anti-HER2 treatment. The study investigated the addition of evorpacept, a next-generation CD47 inhibitor, to the standard combination of trastuzumab, ramucirumab, and paclitaxel (TRP). Results showed that adding evorpacept led to a higher objective response rate compared to TRP treatment alone. It was found that the treatment response was associated with CD47 and HER2 expression.
